Researchers have analyzed data from more than one million participants in the largest study of human behavioral genetics to date, uncovering 1,260 common genetic variants linked to the “Big Five” personality traits. Published in Nature, the findings establish biological links between genetic architecture and lifestyle outcomes such as smoking habits and infection risks.
Uncovering the Genetic Architecture of the Big Five Personality Traits
An international research effort led by the University of Bristol has mapped the genetic underpinnings of personality on an unprecedented scale. By pooling data from 46 cohorts representing between 611,000 and 1.14 million individuals through the Revived Genomics of Personality Consortium, scientists examined the cellular and genetic basis of the Big Five personality model: extraversion, agreeableness, conscientiousness, neuroticism, and openness to experience.
The study identified 1,260 common genetic variants associated with one or more of these core traits, including more than 800 variants that had not been previously documented according to the Nature report. The genome-wide association study, or GWAS, substantially increased the number of known genetic markers tied to specific psychological profiles. For conscientiousness, known variants rose from 3 to 131, while genetic variants linked to neuroticism jumped from 224 to 706. Across the participant pool, common genetic variants accounted for between 5% and 16% of the variation in the Big Five traits depending on the specific characteristic and analytical variables as detailed in the published data.
Independent of Family Environment and Childhood Upbringing
A central question in behavioral genetics has been whether family background or early childhood environments shape the genetic correlations observed in adulthood. To investigate this, researchers conducted separate analyses involving related participants, including twin comparisons and cross-generational evaluations between parents and siblings reported by the international team.

The statistical models demonstrated that the genetic variants influencing personality operate largely independently of shared household environments or early upbringing according to lab and statistical evaluations.
Michel Nivard, genetic epidemiologist at the University of Bristol, noted that he had figured there would be some of that going on, some indirect genetic effects, where an early environment shaped by the genes in one’s parents would influence personality, but added that they simply did not find that kind of gene–environment correlation.
Connecting Personality Variants to Health and Lifestyle Habits
By utilizing these genetic correlations, researchers established statistical bridges between innate psychological traits and real-world health outcomes. Meanwhile, markers for openness to experience tracked with preferences for nocturnal activity and specific dietary tastes including coffee, wine, and aged cheese highlighted in the primary journal documentation.

Behavioral geneticists emphasize that these insights serve as tools for understanding the biological architecture underlying behavioral and mental health outcomes rather than serving as diagnostic instruments for individuals. Researchers note that while the field of personality genetics has historically lagged behind studies of other behavioral traits as observed by Aysu Okbay of Amsterdam University Medical Centre, the current dataset opens a new phase in social genomics according to study co-leader Michel Nivard.
Nivard added in interviews detailing the methodology that he was not in the business, and had no interest at all, in predicting personality, because if he wanted to know someone’s personality, he would simply measure it, stressing that the genome functions primarily as a research instrument to investigate human differences rather than an individual forecasting tool.
